The rich nutritional base of TSB, supplemented with SPS and CO2, is an excellent broth for blood cultures. TSB conforms to Harmonized United States Pharmacopoeia (USP), European Pharmacopoeia (EU), and Japanese Pharmacopoeia (JP). TSB was chosen by the USDA Animal and Plant Health Inspection Service for detecting viable bacteria in live vaccines. TSB is recommended for testing bacterial contaminants in cosmetics and complies with established standards in the food industry. Clostridia and non-sporulating anaerobes grow luxuriantly in this broth when incubated under anaerobic conditions. This medium was originally developed for use without blood in determining the effectiveness of sulfonamides against pneumococci and other organisms. Tryptic Soy Broth, a general purpose medium, is commonly referred to as Casein Soya Bean or Soybean-Casein Digest broth, and abbreviated as TSB. Tryptic Soy Broth conforms to Harmonized USP/EP/JP requirements and is not intended for use in the diagnosis of disease or other conditions in humans. Tryptic soy agar containing various salt concentrations is also available which are recommended for use in determining the halotolerant levels of microorganisms.Tryptic Soy Broth (Soybean-Casein Digest Broth) is used for the cultivation of a wide variety of microorganisms.

Tryptic soy agar can be used in determining the X, V, and XV factors requirements of Haemophilus species by using strips or discs containing X, V, and XV factors in the inoculated plates. 5% sheep blood), TSA may be used as a primary isolation medium in clinical microbiology. TSA is not used for the isolation of pathogens from clinical specimens but may be used for maintaining or subculturing bacterial strains (e.g., Enterobacteriaceae and Staphylococci). Use of TSA in clinical microbiology is limited as it does not support the growth of a variety of fastidious bacteria. This medium is recommended for use in the cultivation, storage, maintenance, and transportation of pure cultures of microorganisms.

Tryptic soy agar supports the growth of nonfastidious as well as moderately fastidious microorganisms. Tryptic soy agar (TSA) is recommended for use as a general growth medium for the isolation and cultivation of microorganisms. Tryptic Soy Agar plate (source:mkldiagnostics) Usage of Tryptic Soy Agar (TSA) Pre-warm the medium to room temperature before usage. Fully cool autoclaved tubes at room temperature before placing stock in 4☌ refrigerator and avoid light.Remove the tubes and make sure the cap of tube is closed tightly. After aliquot, place all tubes into autoclave at 121☌ for 15 minutes.Aliquot 10 ml of the medium to each 13*100 mm glass spiral tubes (or other suitable containers as per your need).Place the bottle on magnetic stirrer to mix.Weight 3 gm of Tryptic soy broth (TSB) powder and dissolve in 100 ml distilled water.Sodium chloride is added to maintain the osmotic equilibrium and agar is the solidifying agent. The combination of casein and soy peptones renders the medium nutritious by supplying organic nitrogen, particularly amino acids and longer-chained peptides. Tryptic soy agar contains digests of casein and soybean meal. Usage of Tryptic Soy Agar (TSA) Composition of Tryptic Soy Agar (TSA) Ingredients Amount (gm/L) Pancreatic digest of casein 15.0gm Peptic digest of soybean meal 5.0gm Sodium chloride 5.0gm Agar 15.0gm Distilled water 1 liter.
